A common way is to deprotonate a molecule, such as $E t O H \to E t O^{-} + H^{+}$ $EtOH -> EtO^(-) + H^(+)$

where $E t O H = C H_{3} C H_{2} O H$ $EtOH = CH_3CH_2OH$

A more interesting way is to put a molecule in solution with an alkali salt such as $L i C l$ $LiCl$ , so something like butyllithium is a great nucleophile.
