Understand that BOTH have constraints on statistical validity.

Spectrophotometry may be faster, which has some advantages in some studies, but it must be calibrated and still has statistical issues with sample size and content.

Plating takes longer, but can be redone as often as desired – both replicate samples and recounting a particular sample. Again, sample size and conditions may affect the statistical validity of the results.

But, for the highest level of statistical significance and the most scientifically rigorous requirements for recounting or comparison, plating is the better method.
