DNA methylation affects the regulation of transcription in that it sometimes allows chromatin to be unwound from histones, and hence be open for transcription.
However, it can also sometimes act as an inhibitor, and inhibit the unwinding of the chromatin from the histones. This does not allow transcription to occur.
